Goat Anti-DGAT1 (Diacylglycerol O-acyltransferase 1, ACAT-related Gene Product 1, Diglyceride Acyltransferase, AGRP1, DGAT)

Glucose and insulin are anabolic signals which upregulate the transcriptions of a series of lipogenic enzymes to convert excess carbohydrate into triglycerides for efficient energy storage. DGAT1 is a microsomal enzyme that assists in the synthesis of fatty acids into triglycerides. DGAT catalyzes the terminal and only committed step in triacylglycerol synthesis by using diacylglycerol (DAG) and fatty acyl CoA as substrates. DGAT plays a fundamental role in the metabolism of cellular diacylglycerol and is important in higher eukaryotes for physiologic processes involving triacylglycerol metabolism such as intestinal fat absorption, lipoprotein assembly, adipose tissue formation, and lactation. DGAT1 is involved in fat absorption in the intestine and in basal level triglyceride synthesis in adipose tissue, where it is more highly expressed.

Applications
Suitable for use in Immunofluorescence, Flow Cytometry, Immunohistochemistry, ELISA and Western Blot. Other applications not tested.
Recommended Dilution
ELISA: 1:64,000 Western Blot: 0.3-1ug/ml, observed in human and rat duodenum lysates on ~55kD bands Immunohistochemistry: 3.75ug/ml in Paraffin embedded Human Colon. Flow Cytometry: 10ug/ml using A431 cells Immunofluorescence: 10ug/ml, shows strong expression of the protein seen in the endoplasmic reticulum and cytoplasm of A431 cell Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Synthetic peptide corresponding to C-QNSMKPFKDMDYS, from DGAT1. at the internal region of human DGAT1. Species sequence homology: canine, bovine and rat.
Form
Supplied as a liquid in Tris saline, 0.02% sodium azide, pH 7.3, 0.5% BSA.
Purity
Purified by immunoaffinity chromatography.
Specificity
Recognizes human DGAT1. Species Crossreactivity: mouse.
