Mouse Anti-Dynein, Cytoplasmic, 74kD Intermediate Chains (MaxLight 650)

MaxLight™650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000.

Applications
Suitable for use in Immunocytochemistry, Western Blot and Immunoprecipitation. Other applications have not been tested.
Recommended Dilution
Immunocytochemistry: Reacts with cultured MDCK cells, NRR cells, N1E cells and PTK-1 cells. Western Blot: Reacts with cultured MDCK cells, PC-12 cells, N1E cells, neurons, glia and other cultured cells. Immunoprecipitation: immunoprecipitates the entire dynein complex stoichiometrically from TX-100 or NP-40 lysates (including the 530kD heavy chain, the light intermediate chains and the light chains) from various tissues and cultured cell lines. If detergents such as SDS are used to prepare the lysates the only dynein subunits which are immunoprecipitated are the IC74 subunits (presumably because the dynein complex dissociates), and other contaminating proteins can be found in SDS immunoprecipitates. Optimal dilutions to be determined by the researcher.
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.

Specificity
Recognizes all known forms of the Mr 74kD intermediate chain subunit (IC74) of mammaliam cytoplasmic dynein. The antigenic site has been localized to the first 60 amino acids of the amino terminal portion of the molecule (a region conserved in both IC74 genes). Species Crossreactivity: Rat, mouse, bovine, sheep, fish, Drosophila and Xenopus. Does not react with squid.
