ERK (extracellular signal regulated kinase), also known as MAPK (mitogen-activated protein kinase) has two closely related isoforms of 44kD and 42kD, respectively. These kinases belong to a family of serine/threonine kinases that are activated upon treatment of cells with a large variety of stimuli including mitogens, hormones, growth factors, cytokines and bioactive peptides. Cell stimulation induces the activation of a signaling cascade, the downstream effects of which have been linked to the regulation of cell growth and differentiation as well as the cytoskeleton. ERK1 and ERK2 are phosphorylated within the activation loop on both a Threonine and a Tyrosine residue (within a Thr-Glu-Tyr motif) by MEKs (MAPK/ERK kinases), thereby greatly elevating the activity of ERK1&2.

Applications
Suitable for use in Western Blot. Other applications not tested.
Recommended Dilution
Western Blot: 1:1000 Optimal dilutions to be determined by the researcher.
Positive Control
A431, PC12 and NIH3T3 cells.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Synthetic peptide corresponding to aa317-339 within the carboxyl-terminal half of the human ERK1 protein. Species sequence homology: human and rat.
Form
Supplied as a liquid in PBS (without Mg2+ and Ca2+), 1mg/ml BSA (IgG, protease free), pH 7.2, 0.05% sodium azide, 50% glycerol.
Purity
Purified by immunoaffinity chromatography.
Specificity
Recognizes human, mouse, rat and bovine ERK1&2. Other species have not been tested.
