Mouse Anti-FK506 Binding Protein 8 (FKBP8, FK506 Binding Protein 38kD, FKBP38, Rotamase)
FK506 Binding Proteins (FKBPs) are intracellular receptors for the immuno-suppressive drug FK506. The FKBP/FK506 complex exerts its immunosuppressive effects by inhibiting calcineurin, a calcium- and calmodulin-dependent serine/threonine phosphatase that functions as a critical signaling molecule during T-cell activation. FKBP38, also known as FKBP8, is a 355aa protein with a calculated molecular weight of 38.7kD and an apparent molecular mass of ~60-64kD in SDS-PAGE. FKBP38 binds to and inhibits calcineurin even in the absence of FK506, indicating that FKBP38 is a constitutively active inhibitor of calcineurin. Additionally, FKBP38 co-immunoprecipitates with Bcl-2 and Bcl-xL, suggesting that FKBP38 may regulate apoptosis by anchoring Bcl-2 and Bcl-xL to mitochondrial membranes. Mouse FKBP38 shares 94% and 87% sequence identity with human and rat FKBP38, respectively.

Recommended Dilution
Western Blot: 0.5ug/ml using HepG2 human hepatocellular carcinoma cell line, NIH-3T3 mouse embryonic fibroblast cell line and L6 rat myoblast cell line Optimal dilution to be determined by researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recombinant protein corresponding to Met58-Gly382 from human FKBP38, expressed in E. coli.

Supplied as a lyophilized powder from PBS, 5% trehalose. Reconstitute with 200ul sterile PBS.
Purity
Purified by Protein G affinity chromatography from hybridoma culture supernatant.
Specificity
Recognizes endogenous human FKBP38 on Western Blots. Species Crossreactivity: mouse and rat.
