Rabbit Anti-GABA Transporter 2 (GAT-2, (Gamma aminobutyrIc acid)

GABA is a major inhibitory neurotransmitter and the GABAergic transmission is terminated by the rapid Na+/Cl-dependent uptake of through GABA transporters. It has been subdivided into neural and glial uptake systems on the basis of pharmacological properties. Recently, molecular cloning studies have identified multiple subtypes of GABA transporters (GAT1, GAT2, GAT3; and betaine GABA transporter (BGT-1). There is ~50% homology between various GABA transporter subtypes. GABA transporters are predicted to contain 12 potential transmembrane domains. The NH2 and COOH-termini are predicted to be intracellular. Two of the high affinity (Km~10 uM) rat GABA transporters (GAT2 and GAT3/GAT-B) share higher amino acid identity (68% and 65%, respectively) with the kidney betaine transporter than with GAT-1 (52% AA identity). GAT1 and GAT3 have been detected in various parts of the brain while GAT2 is found in many tissues. It appears that GAT1 and GAT3 are involved in distinct GABAergic transmission while GAT2 may be important in non-neural functions.

Applications
Suitable for use in Immunohistochemistry and Western Blot. Other applications have not been tested.
Recommended Dilutions
Immunohistochemistry: 1:500-1:750 on 4% paraformaldehyde fixed tissue. It is recommended that the antibody is diluted in PBS containing 10% normal goat serum and 0.5% Triton X-100. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Mouse GAT-2, C-terminus (aa593-602).

Supplied as a lyophilized powder from 0.1M PBS, 0.09% sodium azide. Reconstitute with 20ul sterile ddH2O.
Purity
Purifed by affinity chromatography.
Specificity
Recognizes the C-terminus of mouse GABA Transporter-2 (GAT-2). No cross-reactivity to the C-termini of other transmitter transporters detected. Species crossreactivity: rat.
