Germinal center kinase (GCK) is the founding member of the GCK family, a group of serine/threonine kinases homologous to the yeast Ste20 kinase. GCK is activated by TNF and associated with TRAF2 (TNF receptor-associated factor 2) and MEKK1, thereby activating the SAPK/JNK pathway (1,2). GCK does not significantly phosphorylate MEKK1, instead it enhances MEKK1 oligomerization and autophosphorylation (3). GCK binds and activates MLK3, indicating that GCK may have other effectors (3).
Applications
Suitable for use in Western Blot. Other applications not tested.
Recommended Dilutions
Western Blot: 1:1000. Incubate membrane with diluted primary antibody in 1X TBS,5% BSA, 0.1% Tween at 4°C with gentle shaking, overnight. Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Synthetic peptide corresponding to residues surrounding Ser380 of human GCK.

Supplied as a liquid in 10mM sodium HEPES, pH 7.5, 150mM sodium chloride, 0.1mg/ml BSA, 50% glycerol.
Purity
Purified by Protein A and peptide affinity chromatography.
Specificity
Recognizes endogenous levels of total human GCK protein at 91kD. Does not crossreact with HPK or other members of the GCK family. Species Crossreactivity: mouse, rat, monkey.
