Mouse Anti-GluR3 (Glutamate Receptor) (MaxLight 490)

MaxLight™ 490 is a new Blue-Green photostable dye conjugate comparable to DyLight™488, Alexa Fluor™488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm); Emission (515nm); Extinction Coefficient 73,000.

Glutamate is the major excitatory neurotransmitter in the mammalian central nervous system. Regulation of fast excitatory synaptic neurotransmission is mediated primarily by ionotropic glutamate receptors (iGluR); ligand-gated cation (principally Ca2+) channels found on postsynaptic neurons throughout the central nervous system. Pharmacologic properties and amino acid sequence similarities allow classification of iGluR into three main subfamilies named for their principal agonists: 1) NMDA (N-methyl-D-aspartate) receptors, and the two non-NMDA receptor subfamilies, 2) AMPA (a-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid) receptors and 3) kainate. iGluR are oligomeric protein complexes comprised of 4 or 5 subunits, with the subunit composition determining specific electrophysiological properties. All NMDA receptors contain one NR1 subunit (encoded by a single gene producing 8 identified splice variants) and several NR2 subunits (four genes, A-D). AMPA and kainate receptors are comprised of subunits GluR1-4 (AMPA) and subunits GluR5-7, KA1 and KA2 (kainate).
Applications
Suitable for use in Western Blot, Immunocytochemistry, Immunofluorescence and Electron Microscopy. Other applications have not been tested.
Recommended Dilutons
Optimal dilutions to be determined by the researcher.
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™490 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.

Purified by Protein A affinity chromatography.
Specificity
Recognizes rat Glutamate Receptor 3 (GluR3) at ~98kD. Does not react with cells transfected with GluR1, GluR2 or GluR4. Species Crossreactivity: human and monkey. Reactivity has been confirmed with HEK293 cells transfected with AMPA GluR cDNAs as well as human, rat, and macaque monkey brain extracts by Western Blot and rat hippocampus by immunofluorescence, immunocytochemistry, and immunogold labeling/electron microscopy.
