Rabbit Anti-Glutamate Dehydrogenase (Biotin)
GDH, GLDH, GLUD 1, GLUD, GLUD1, GLUD1, Glutamate dehydrogenase 1, Glutamate dehydrogenase 1 mitochondrial, MGC132003
Glutamate is a major excitatory neurotransmitter. One enzyme central to the metabolism of glutamate is glutamate dehydrogenase GDH1, that catalyzes the reversible deamination of L-glutamate to 2-oxoglutarate using NAD+ or NADP+. Mammalian GDH is composed of six identical subunits, and the regulation of GDH is very complex. It has been a major goal to identify the substrate and regulatory binding sites of GDH. It is only in recent years that the three-dimensional structure of GDH from microorganisms is available. Very recently, crystallization of bovine liver GDH was reported for the first time from the mammalian sources. However, remarkably little is known about the detailed structure of mammalian GDH, especially the brain enzymes. Bovine glutamate dehydrogenase exists as a homohexamer located within the mitochondrial matrix.
Applications
Suitable for use in Immunoblot, ELISA, Immunohistochemistry, Immunomicroscopy. Other applications have not been tested.
Recommended Dilution
ELISA: 1:1000-1:4000. Capture ELISA using peroxidase-conjugated streptavidin, 30 min., RT. Optimal dilutions to be determined by the researcher.
Label: Biotinamidocaproate N-Hydroxysuccinimide Ester (BAC) Biotin/Protein Ratio: 10-20 BAC molecules per rabbit IgG molecule
Storage and Stability
May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Bovine liver glutamate dehydrogenase.
Form
Supplied as a liquid in PBS, pH 7.2, 0.01% sodium azide, 40% glycerol. Labeled with Biotin.
Purity
Purified by delipidation, fractionation and ion exchange chromatography.
Specificity
Recognizes Glutamate Dehydrogenase. Assay by immunoelectrophoresis resulted in a single precipitin arc against bovine liver glutamate dehydrogenase, anti-biotin and anti-rabbit serum. Crossreactivity from other tissues and species may occur but have not been determined.
