Mouse Anti-Hepatitis C Virus NS4 (HCV)
Hepatitis C virus (HCV) causes chronic hepatitis and liver cirrhosis in human through blood and body fluid transmission. HCV has a positive sense single RNA genome enclosed in the nucleocapsid made of core protein (capsid protein). The nucleocapsid is covered by an envelope made of lipoproteins (E1 and E2). The 9.6Kb HCV genome has a single open-reading frame, which is to be translated into a single polyprotein. HCV viral proteins are produced after processing the polyprotein. Genes for core protein and envelop proteins are located adjacently at the 5’-end of HCV genome, followed by genes for non- structural proteins including NS2, NS3, NS4A, NS4B, NS5, NS5A and NS5B.
Applications
Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilutions
Western Blot: 0.5ug/ml will alow detection of 0.5ug/lane of synthetic NS-4 peptide and 0.1ug/lane of recombinant chimeric HCV polyprotein. Immunohistochemistry: Formalin-fixed paraffin embedded tissues using ABC Optimal dilutions to be determined by the researcher.
Hybridoma
Sp2/0-Ag14 myeloma cells with spleen cells from Balb/c mice.
Storage and Stability
Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Synthetic polypeptide in which overlapping sequences of at least 90 residues in length were selected from the immunodominant NS4 regions of Chinese HCV strains.
Form
Supplied as a lyophilized powder from PBS, pH 7.0. Reconstitute with 500ul sterile ddH2O.
Purity
Purified by Protein G affinity chromatography.
Specificity
Recognizes synthetic human NS4a Protein and Recombinant Chimeric HCV Polyprotein. No crossreaction with HCV Capsid region and other non-structure region.
