Mouse Anti-Interleukin 7 Receptor alpha (Interleukin-7 Receptor Subunit alpha, IL-7 Receptor Subunit alpha, IL-7R Subunit alpha, IL-7R-alpha, IL7RA, IL-7RA, IL7R, CD127, CDw127, ILRA)

CD127 (IL-7R alpha) is the specific receptor component for the cytokine Interleukin -7 (IL-7). It is found on a wide variety of hematopoietic cell types including B cell precursors, and the majority of T cells. Its expression levels are decreased on T cells following activation. CD127 can dimerize with CD132 (IL-2R gamma) to form a high affinity IL-7 receptor (1). CD127 engagement is necessary for T cell development in humans (2).

Applications
Suitable for use in Flow Cytometry. Other applications not tested.
Recommended Dilution
Optimal dilutions to be determined by the researcher.
Flow Cytometry Protocol
1. 5x10e5 human Nalm-6 cells were washed incubated 45 minutes on ice with 80ul of I8429-27M1 at 10ug/ml. 2. Cells were washed twice and incubated with IgG (FITC) Goat-anti-Mouse . 3. The cells were washed twice, fixed and analyzed by FACS. Cells stained positive with a mean shift of 0.71 log10 fluorescent units when compared to isotype control Mouse IgG1 antibody. Binding was blocked when reagent was pre-incubated with a 10-fold excess of recombinant CD127-muIg (cat# I8429-27N).
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recombinant correspponding to human CD127.
Form
Supplied as a liquid in 50mM sodium phosphate, pH 7.5, 100mM potassium chloride, 150mM sodium chloride, 0.5mg/ml gentamicin sulfate.
Purity
Purified by Protein A affinity chromatography.
Specificity
Recognizes an extracellular epitope of recombinant and native human CD127 in FACS and ELISA.
