Mouse Anti-Mast Cell Chymase (CMA1)

Chymase-1 (also mast cell protease I) is a secreted, 30kD glycoprotein member of the granzyme subfamily, peptidase S1 family of enzymes. It is secreted by mast cells and vascular smooth muscle cells, and converts angiotensin I into angiotensin II. This promotes fibroblast proliferation and secretion of ECM. Mature human chymase is 226aa in length. It contains one peptidase S1 domain (aa 2-245). There is one potential alternate splice form. It shows a deletion of the 99 C-terminal amino acids (aa149-247).

Applications
Suitable for use in Direct ELISA, Western Blot and Immunoprecipitation. Other applications not tested.
Recommended Dilutions
Western Blot: 1ug/ml using human small intestine tissue Immunoprecipitation: 25ug/ml using conditioned cell culture medium spiked with recombinant human Chymase/CMA1 Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Supplied as a lyophilized powder from PBS, 5% trehalose. Reconstitute with 200ul sterile PBS.
Purity
Purified by Protein A affinity chromatography from hybridoma culture supernatant.
Specificity
Recognizes human Chymase at ~30kD. No cross reactivity with rmMCPT-1, rmMCPT-6, rmMCPT-7 or rmMCPT-11.
