Mouse Anti-Matrix Metalloproteinase 13 (MMP-13, Collagenase-3)

Human collagenase-3 (MMP13) is a recently identified member of the matrix metalloproteinase (MMP) family that is expressed in breast carcinomas and in articular cartilage from arthritic patients. The MMP-13 gene has been isolated and characterized. This gene is composed of 10 exons and 9 introns and spans over 12.5 kb. The overall organization of the collagenase-3 gene is similar to that of other MMP genes clustered at chromosome 11q22, including fibroblast collagenase (MMP-1), matrilysin (MMP-7), and macrophage metalloelastase (MMP-12), but is more distantly related to genes coding for stromelysin-3 (MMP-11), gelatinase-A (MMP-2), and gelatinase-B (MMP-9), which map outside of this gene cluster.

Applications
Suitable for use in Immunohistochemistry and Immunoprecipitation. Other applications have not been tested.
Recommended Dilution
Immunohistochemistry (Frozen and formalin-fixed/paraffin): 5-10ug/ml for 30-60 minutes at RT. Staining of formalin-fixed tissues requires boiling of tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 minutes followed by cooling at RT for 20 minutes. Immunoprecipation: (Native verified) 2ug/mg protein lysate. Use Protein G. Optimal dilutions to be determined by the researcher.
Positive Control
Conditioned, serum-free medium from (TPA-treated) human chondrosarcoma SW1353 cells. Placenta, breast, ovarian carcinomas.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Supplied as a liquid in 10mM PBS, pH 7.4, 0.2% BSA, 0.09% sodium azide.
Purity
Purified by Protein G affinity chromatography from ascites.
Specificity
Recognizes human MMP-13 at ~60kD and ~48kD which are identified as pro (latent) and active forms of matrix metalloproteinase-13. Shows no crossreactivity with the pro and active forms of other MMPs. Species Crossreactivity: rabbit and mouse.
