Rabbit Anti-Melanoma Inhibitory Activity Protein (MIA)

MIA acts as a potent tumor cell growth inhibitor for malignant melanoma cells and some other neuroectodermal tumors, including gliomas, in an autocrine fashion. In a study of human melanoma cell lines with different metastatic capacity MIA mRNA expression appeared to be inversely correlated with pigmentation. MIA has been shown to represent a very sensitive and specific serum marker for systemic malignant melanoma that might be useful for staging of primary melanomas, detection of progression from localized to metastatic disease during follow-up, and monitoring therapy of advanced melanomas.

Applications
Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilutions
ELISA (Sandwich): 0.5-2ug/ml using 100ul/well antibody solution in conjunction with biotinylated anti-human Melanoma Inhibitory Activity Protein (M2869-95B) as a detection antibody, allows the detection of at least 2000-4000pg/well of recombinant human Melanoma Inhibitory Activity Protein. Western Blot: 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant human Melanoma Inhibitory Activity Protein is 1.5-3ng/lane, under either reducing or non-reducing conditions. Immunohistochemistry (FFPE): 1-2ug/ml using human pancreas infiltrating ductal adenocarcinoma tissue with an overnight incubation at 4ºC. An HRP-labeled polymer detection system was used with an alcohol-soluble AEC chromogen. Optimal results for these conditions were acheived with heat induced antigen retrieval with sodium citrate, pH 6 or enzyme induced antigen retrieval with proteinase K at RT. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recombinant protein corresponding to human Melanoma Inhibitory Activity Protein expressed in E. coli.
Form
Supplied as a lyophilized powder from PBS, pH 7.2. Reconstitute with sterile ddH2O to a concentration of 0.1-1mg/ml.
