Mouse Anti-Mitochondria

This antibody recognizes a 60kD nonglycosylated protein component of mitochondria found in human cells. It can be used to stain mitochondria in cell, tissue or biochemical preparations and can be used as a marker of the mitochondria in subcellular fractions.

Applications
Suitable for use in Indirect Immunofluorescence, Immunohistochemistry, and Immunoprecipitation. Can also be used for isolation of intact mitochondria from freeze/thaw lysates of living cells via conjugation to magnetic beads. Other applications have not been tested.
Recommended Dilution
Indirect Immunofluorescence: 1:10-1:50, 30-100ul per slide well Immunohistochemistry (paraffin-embedded tissues): 1:80 Using heart ventricle epithelium pretreated with EDTA, pH 8.0. Detection with HRP-DAB. Immunoreactivity is seeing the circular-granular layer surrounding the nucleus of the cells. Requires citric acid/microwave antigen retrieval; enhanced ABC or amplified detection systems recommended. Immunohistochemistry: 1:100. Works best with organic fixatives (acetone, methanol, etc.). 2% PFA for 10-15 minutes. can be used but stronger use of formalin or 4% PFA limits staining. Optimal dilutions to be determined by the researcher.

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Purity
Purified by ammonium sulfate precipitation from hybridoma supernatant.
Specificity
Recognizes human mitochondria. Gives mitochondrial staining on all human cell types. This pattern is best described as a spaghetti- like staining pattern in fibroblasts and a perinuclear large speckled pattern in lymphonoid cells. Recognizes a 65kD protein by immunoprecipitation. Does not cross-react with rat and mouse. Epitope: surface of intact mitochondria.
