Goat Anti-MUCDHL (Mucin and Cadherin-Like Protein, mu-Protocadherin)
Mucin and cadherin-like protein (MUCDHL), also known as μ-protocadherin, is a 110kD type I transmembrane protein that is both O- and N-glycosylated. It is a calcium-dependent cell adhesion protein in the cadherin superfamily. MUCDHL is found on intestinal and renal epithelium. Human MUCDHL extracellular domain (ECD) contains four cadherin domains (residues 71-459) plus several mucin domains (residues 540-645). Alternate splicing creates four isoforms that differ in their mucin domains. In the region used as immunogen, human and mouse MUCDHL share 63% amino acid sequence identity.
Applications
Western Blot: This antibody can be used at 0.1-0.2ug/ml with the appropriate secondary reagents to detect human MUCDHL. The detection limit for rhMUCDHL is approximately 5 ng/lane under non-reducing and reducing conditions. Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect human MUCDHL. The detection limit for rhMUCDHL is approximately 0.3 ng/well. Optimal dilutions should be determined by each laboratory for each application.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
E. coli-derived rhMUCDHL (aa26-434)
Form
Supplied as a lyophilized powder from PBS, 5% trehalose. Reconstitute with 500ul sterile PBS.
Purity
Purified by immunoaffinity chromatography.
Specificity
Selected for its ability to recognize human MUCDHL in direct ELISAs and Western Blots. In these formats, this antibody shows ~10% crossreactivity with rmMUCDHL.
