Myosin heavy chain type A is a critical component of the cellular contractile apparatus. It is involved in maintaining cell-cell adhesion in early mammalian embryos and ablation of this protein leads to prenatal lethality. Mutation in the human MYH9 gene lead to MYH9-related hereditary macrothrombocytopenia (HMTC) characterized by a reduced number of blood platelets.

Applications
Suitable for use in Western Blot, Immunofluorescence and Immunocytochemistry. Other applications not tested.
Recommended Dilutions
Immunofluorescence (IC): 1:50-1:100 Western Blot: 1:1000-1:2000. Detects a band at ~200kD. Staining of myosin IIA (nonmuscle) is inhibited by the immunizing peptide. Additional bands may be detected in some preparations. Optimal dilutions to be determined by the researcher.
Positive Control
MDCK or Jurkat whole cell extract (Western Blot). NRK cells (Immunofluorescence).
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Synthetic peptide corresponding to aa1949-1960 of human non-muscle Myosin IIA, conjugated to KLH.
Form
Supplied as a liquid in 0.01M PBS, pH 7.4, 1% BSA, 15mM sodium azide.
Purity
Purified by affinity chromatography.
Specificity
Recognizes human myosin IIA. Species Crossreactivity: rat and canine
