Mouse Anti-Nkx6.1 (NK Homeobox Factor 6.1, NKX6A)

Nkx6.1 (NK homeobox factor 6.1; also NKX6A) is a 39-43kD member of the homeodomain family of transcription factors. It is expressed in embryonic ventral neural tube where it drives neural progenitors to form motoneurons and V2-type interneurons. It is also expressed in both fetal and neonatal pancreas where it promotes a b-cell phenotype. Nkx6.1 exhibits both transcriptional gene repression (Nkx6.2 and Dbx2) and activation (cyclin A2 and B1). When overexpressed, it induces b cell mitosis. Human Nkx6.1 is 367aa in length. It contains a repressor region composed of Pro and Ala (aa136-173), a DNA binding homeobox (aa236-295) and a Glu:Asp-rich transactivation domain (aa305-337). Over (aa1-120), human Nkx6.1 shares 96% aa identity with mouse Nkx6.1.

Applications
Suitable for use in Direct ELISA, Western Blot and Immunocytochemistry. Other applications not tested.
Recommended Dilutions
Immunocytochemistry: 8-25ug/ml using immersion fixed BTC-6 mouse beta cell insulinoma cell line. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Specificity
Recognizes human Nkx6.1. Does not crossreact with recombinant human Nkx2E, 2.2, 3.1 or recombinant mouse Nkx3.1.
