Rabbit Anti-nNOS (MaxLight 550)

Nitric oxide (NO) is a potent bio-regulatory molecule, which plays important roles in the regulation of a variety of normal developmental and physiological processes. NO is generated by nitric oxide synthase (NOS) in a reaction which converts L-arginine and oxygen into citrulline and nitric oxide. Three distinct mammalian genes have been identified which encode NOS isoforms: neuronal (nNOS or bNOS), macrophage or inducible (iNOS), and endothelial (eNOS). The NOS isoforms can be subdivided into two general categories, constitutive or inducible, based on differences in their regulation and activities. The constitutive isoforms include eNOS and nNOS.These isoforms are always present but remain inactive until an increase in intracellular calcium results in enhanced calcium/calmodulin binding and subsequent activation. In contrast, the inducible NOS isoform, iNOS, is not normally expressed, but is induced (though alterations in gene expression, mRNA stability, or protein synthesis) in response to certain cytokines.

Applications
Suitable for use in FLISA, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilutions
FLISA: 0.1-1.0ug/ml Western Blot: 1ug/ml Immunohistochemistry (Paraffin): 5-10ug/ml. Some background staining is evident in paraffin embedded tissues Immunohistochemistry (Frozen): 1-2ug/ml Optimal dilutions to be determined by the researcher.
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™550 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
Immunogen
A recombinant protein consisting of 195 amino acids from the N-terminal of rat nNOS protein.

Supplied as a liquid in PBS, pH 7.2. No preservatives added. Labeled with MaxLight™550.
Purity
Purified by immunoaffinity chromatography.
Specificity
Recognizes the ~160kD nNOS protein and does not exhibit any crossreactivity with the related eNOS or iNOS proteins. |Species Crossreactivity: Reacts with a ~160kD band on Western Blots of rat and mouse brain tissue lysates (20ug).
