Mouse Anti-p19skp1 (S-phase Kinase-associated Protein) (MaxLight 650)

MaxLight™ 650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000.

The SKP1 gene was first identified in yeast, where it is essential for the G1/S and G2/M cell cycle transitions.1 Its protein product, Skp1 (p19), interacts with cyclin F directly and cyclin A indirectly, and is thought to link these key regulatory proteins of the cell cycle to ubiquitin-mediated degradation by the proteolytic pathway. Skp1 is a subunit of a type of E3 ubiquitin ligase known as an SCF ligase, for its Skp1, Cul1, and F-box protein components. Within the SCF complex, Cul1 functions as the catalytic core which recruits the E2 ubiquitin-conjugating enzyme, the F-box protein provides substrate specificity for the protein to be degraded,2 and Skp1 acts as an adapter protein linking the F-box protein to Cul1.3 De-regulation of SCF-dependent protein degradation has been implicated as a contributing factor in malignant transformation. The complete loss or increased proteolysis of cyclin E, b-catenin, and p27, all of which are degradation targets of specific SCF ligases, has been observed as a frequent event in cancer.3 The human SCF ligase containing Skp2 as its F-box protein (SCFSkp2) degrades p27, SCFTrcp targets b-catenin,4 and SCFFbw7/hCdc4/hAgo degrades cyclin E.
Applications
Suitable for use in FLISA, Western Blot and Immunoprecipitation. Other applications not tested.
Recommended Dilutions
FLISA (Immunogen): 0.1-1.0ug/mL Western Blot: 1-3ug/ml Immunoprecipitation: 5-10ug/ml Optimal dilutions to be determined by the researcher.

Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. 
Note: Applications are based on unconjugated antibody.
