Phosphoprotein Associated with Glycosphingolipid Microdomains 1

PAG (phosphoprotein associated with GEMs), also known as Cbp (Csk-binding protein), is a ubiquitously expressed 46kD transmembrane adaptor protein present in membrane rafts (glycosphingolipid-enriched microdomains), which however migrates on SDS PAGE gels anomalously as an 80kD molecule. Following tyrosine phosphorylation by Src family kinases, PAG binds and thereby activates the protein tyrosine kinase Csk, the major negative regulator of the Src family kinases. Signaling via the B-cell receptor in B cells or high affinity IgE receptor (FcepsilonRI) in mast cells leads to PAG increased tyrosine phosphorylation and Csk binding, while T cell receptor signaling causes PAG dephosphorylation, loss of Csk binding and increased activation of the protein tyrosine kinase Lck.

Applications
Suitable for use in Western Blot, Flow Cytometry and Immunohistochemistry. Other applications not tested.
Recommended Dilutions
Flow Cytometry: 2ug/ml. Intracellular staining. Immunohistochemistry (paraffin): Positive tissue: tonsil, spleen. Western Blot: Csk binding protein is an ubiquitously expressed 46kD transmembrane adaptor protein present in membrane microdomains (rafts), which, however, migrates on SDS-PAGE gels anomalously as an 80kD molecule. Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recombinant intracellular fragment corresponding to aa97-432 from human Cbp (PAG).

Purified by Protein A affinity chromatography.
Specificity
Recognizes an epitope (aa235-280) of Csk-binding protein (Cbp) located in the cytoplasmic domain. Does not react with mouse, rat or bovine
