Applications
Suitable for use in ELISA (direct), Flow Cytometry, Immunohistochemistry and Western Blot. Other applications not tested.
Recommended Dilutions
Flow Cytometry: 2.5ug per 10e6 cells; mouse splenocytes treated with PHA. Immunohistochemistry: 5-15ug/ml using perfusion-fixed frozen sections of mouse thymus. Western Blot: 0.2ug/ml with lysates of EL-4 mouse lymphoblast cell line and CTLL-2 mouse cytotoxic T cell line (negative control) Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Recombinant protein corresponding to Leu25-Gln167 from mouse PD-1, expressed in Sf21 cells (Q02242).
Form
Supplied as a powder lyophilized from PBS, pH 7.4, 5% trehalose. Reconstitute with 500ul sterile PBS.
Purity
Purified by immunoaffinity chromatography.
Specificity
Recognizes mouse PD-1. Shows ~50% crossreactivity with recombinant human PD-1.
