Mouse Anti-Phospholipase C, gamma 1 (PLCg1)

Phospholipase C gamma 1 is a tryosine kinase substrate for many receptor and nonreceptor tyrosine kinases. Activation of the enzyme produces two second messenger molecules, inositol 1,4,5-triphosphate and diacylglycerol, which provoke the mobilization of intracellular Ca2+ and activation of protein kinase C. Although the mammalian genome encodes 10 known phosphoinositide specific PLCs, only the gamma 1 and gamma 2 isoforms are regulated by tyrosine kinase activity. PLC beta isoform activity is controlled by heterotrimeric G protein coupled receptors, while the mechanism of regulation of PLC gamma activity is unknown.

Applications
Suitable for use in ELISA, Immunoprecipitation, Western Blot and Immunocytochemistry. Other applications have not been tested.
Recommended Dilution
Western Blot: 0.5-2ug/ml detects PLCg1 in human A431 carcinoma cells as well as rabbit and bovine brain cytosol preparations. Immunoprecipitation: 4ug Immunoprecipitates PLCg1 from non-stimulated A431 cell lysates and from rabbit brain cytosol preparation. Immunocytochemistry: 5-10ug/ml detects PLCg1 on ethanol-acetic acid (95:5) fixed A431 cells. Optimal dilutions to be determined by the researcher.
Positive Antigen Control (Included)
A0001-50 (Non-stimulated A431 cell lysate). Add 2.5ul of 20mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Use 20ug per lane for minigels.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Five individual clones raised against purified soluble phospholipase C (PLC) g-1 from bovine brain.
Form
Supplied as a lyophilized powder from 0.1M Tris-glycine, pH 7.4, 0.15M sodium chloride, 0.05% sodium azide. Reconstitute with 40ul dH2O.
Purity
Purified by Protein G affinity chromatography.
Specificity
Recognizes bovine PLC g-1 at ~135kD. Reacts with some non-PLC g-1 SH-3 containing proteins, including nck (47kD). Species crossreactivity: rabbit, rat, mouse and human.
