Mouse Anti-Pregnancy Associated Plasma Protein A (PAPP-A, Pappalysin 1, PAPA, PAPPA, PAPP-A, PAPPA1, Aspecific BCL2 ARE-binding Protein 2, ASBABP2, Differentially Placenta 1 Expressed Protein, DIPLA1, EC 3.4.24.79, IGF-dependent IGFBP-4 Protease, IGFBP-4ase, Insulin-like Growth Factor-dependent IGF-binding Protein 4 Protease, Pappalysin 1 Precursor) (FITC)

PAPP-A was initially identified as a high molecular mass component in human pregnancy serum. In pregnancy serum PAPP-A exists in a disulfide bound heterotetrameric complex consisting of two 200kD PAPP-A subunits and two 50-90kD subunits of the proform of eosinophil major basic protein (proMBP). The concentration of PAPP-A in serum increases with gestational age, however, in Down’s syndrome pregnancies PAPP-A concentration is markedly reduced in the first trimester. As such, PAPP-A is widely used as an early marker for Down’s syndrome. PAPP-A has also been shown to exist in a homodimeric, 400kD form in unstable atherosclerotic plaques. Increased circulating concentrations of this dimeric PAPP-A are associated with acute coronary syndromes.

Applications
Detection of human PAPP-A antigen in FLISA, Western Blot. Other applications not tested.
Recommended Dilution
Optimal dilutions to be determined by the researcher.
Matched Pairs: for the detection of PAPP-A in blood of AMI patients and patients with unstable angina (Capture-Detection): P6000-84 – P6000-86, P6000-84 – P6000-87, P6000-82A – P6000-80A
Matched Pairs: for the detection of PAPP-A in pregnancy serum (Capture – Detection): P6000-84 – P6000-86, P6000-84 – P6000-87, P6000-85 – P6000-86, P6000-82A – P6000-80A, P6000-82A – P6000-81A, P6000-81A – P6000-82A
Hybridoma
Derived from hybridization of SP2/0 myeloma cells with spleen cells of Balb/c mice immunized with purified human PAPP-A/proMBP complex.
Storage and Stability
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
Immunogen
Purified human PAPP-A/proMBP complex
Form
Supplied as a liquid in PBS, pH 7.2. No preservative added. No stabilizing proteins added. Labeled with Fluorescein isothiocyanate (FITC).
Purity
Purified by Protein A affinity chromatography.
Specificity
Human PAPP-A antigen purified from placental blood (heterotetrameric complex, consisting of two PAPP-A subunits and two pro-MBP subunits). Recognizes PAPP-A subunit in the heterotetrameric complex in Western Blot.

Intended for research use only. Not for use in human, therapeutic, or diagnostic applications.
