Rad51 is a structural and functional homolog of Escherichia coli RecA, which is required for genetic recombination, DNA repair, and cellular proliferation. This 43kD protein plays an important role in the repair of DNA double-strand breaks caused by ionizing radiation or from the breakdown of stalled replication forks. It forms helical nucleoprotein filaments in which the DNA lies extended and underwound. Filaments form preferentially on tailed duplex DNA substrates at the chromosomal break sites. Rad51 expression is increased in transformed cells and in cancer cell lines. Rad51 is overexpressed in breast carcinomas. Aberrant overexpression of Rad51 is directly correlated with radiation resistance and chemotherapeutic drug resistance. This aberrant expression of Rad51 is also related to enhanced recombination and genomic instabilities in breast carcinoma cells.
Applications
Suitable for use in Western Blot and Immunoprecipitation. Other applications have not been tested.
Recommended Dilution
Western Blot: 1-3ug/ml Immunoprecipitation: 10ug/IP Reaction Optimal dilutions to be determined by the researcher
Storage and Stability
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Specificity
This antibody recognizes the human RAD51 protein. It can be used to analyse the role of the hRAD51 protein in genetic recombination. Reactivity is confirmed with Hela cells via Western blot analysis. Some cross reactivity has been observed in calf and pig.
