The 21kD guanine-nucleotide binding proteins (K-Ras, H-Ras, and N-Ras) cycle between active (GTP-bound) and inactive (GDP-bound) forms (1). Receptor tyrosine kinases and G-protein coupled receptors activate Ras, which then stimulates the Raf-MEK-MAPK pathway (2-4). GTPase-activating proteins (GAP) normally facilitate the inactivation of Ras, however in 30% of human tumors point mutations in Ras prevent the GAP-mediated switching off of this pathway (5). The most common oncogenic Ras mutation found in tumors is Glycine 12 to Aspartate (G12D), which prevents Ras inactivation possibly by increasing the overall rigidity of the protein (5,6).
Application
Suitable for use in ELISA and Western Blot. Other applications have not been tested.
Recommended Dilution
Western Blot: 1:1000 (~10blots/100ul). Optimal dilutions to be determined by the researcher.
Positive Contol
HT-29, PAE, NIH-3T3, AR42J, PC12 and C6 cell lysates.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Synthetic peptide corresponding to residues close to the N-terminus of human K-Ras. Species Sequence Homology: Chicken, hamster, Xenopus, zebrafish: 100%.
Form
Supplied as a liquid in 10mM sodium HEPES, pH 7.5, 150mM sodium chloride, 0.1mg/ml BSA, 50% glycerol.
Purity
Purified by Protein A and peptide affinity chromatography.
Specificity
Recognizes endogenous levels of total K-Ras, H-Ras and N-Ras. May also crossreact with R-Ras and M-Ras. Species Crossreactivity: Human, mouse, rat.
