Rabbit Anti-RIP3 (Receptor-interacting Serine/Threonine-protein Kinase 3, RIP-like Protein Kinase 3, RIPK3, Receptor-interacting Protein 3, RIP-3, mRIP3) (HRP)

Certain serine/threonine protein kinases, such as ASK1, RIP, DAP, and ZIP kinases, are mediators of apoptosis. Receptor interacting proteins including RIP and RIP2/RICK mediate apoptosis induced by TNFR1 and Fas, two prototype members in the death receptor family. A novel member in the RIP kinase family was recently identified and designated RIP3. RIP3 contains N-terminal kinase domain but, unlike RIP or RIP2, lacks the C-terminal death or CARD domain. RIP3 binds to RIP and TNFR1, mediates TNFR1 induced apoptosis, and attenuates RIP and TNFR1 induced NF-κB activation. Overexpression of RIP3 induces apoptosis and NF-κB activation. The messenger RNA of RIP3 is expressed in a subset of adult tissues.

Applications
Suitable for use in ELISA, Immunohistochemistry, Immunoprecipitation, Immunofluorescence and Western Blot. Other applications have not been tested.
Recommended Dilutions
Immunohistochemistry (paraffin): Requires antigen retrieval using heat treatment prior to staining of paraffin sections. Western Blot: detects a band of ~56kD in Mouse 3T3 whole cell lysate Optimal dilutions to be determined by the researcher.
Storage and Stability
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
