Goat Anti-Secretory Leukocyte Protease Inhibitor (SLPI)
Human Secretory Leukocyte Protease Inhibitor SLPI is a protein found in various secretory fluids, including parotid secretions, bronchial, nasal and cervical mucus, and seminal fluid. The protein is a potent inhibitor of a variety of proteolytic enzymes, including the neutrophil proteases, elastase and cathepsin G and the pancreatic proteases chymotrypsin and trypsin. SLPI is an important component of the antiprotease defense of tissues bathed by secretory fluids and could be useful as a therapeutic agent for treatment of the proteolytic damage of tissues seen in degenerative and inflammatory diseases.
Applications
Suitable for use in Neutralization, Direct ELISA, Western Blot, and Immunohistochemistry. Other applications not tested.
Recommended Dilution
Neutralization: Measured by its ability to neutralize Recombinant Human SLPI (0.125ug/ml) inhibition of Trypsin (0.05ug/mL) cleavage of the fluorogenic peptide substrate Mca-RPKPVE-Nval-WRK(Dnp)-NH2 (10uM). The Neutralization Dose (ND50) is typically 2ug/ml. Western Blot: 0.1ug/ml. Recombinant Human SLP1 Immunohistochemistry: 5-15ug/ml using immersion fixed paraffin-embedded sections of human lung Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recombinant protein corresponding to human secretory leukocyte protease inhibitor expressed in E. coli.
Form
Supplied as a lyophilized powder from PBS, pH 7.4, 5% trehalose. Reconstitute with 200ul sterile PBS.

Purified by immunoaffinity chromatography.
Specificity
Recognizes human Secretory Leukocyte Protease Inhibitor in direct ELISA and Western Blot. In direct ELISA less than 15% crossreactivity with mSLPI. Antibody neutralizes SLPI biological activity.
