Rabbit Anti-SGK (SGK1, Serum and Glucocorticoid-regulated Protein Kinase)

The catalytic domain of serum and glucocorticoid-regulated protein kinase (SGK) is 54% identical with that of Akt/PKB. Although SGK1 lacks the PtdIns(3,4,5) P3-binding Pleckstrin-homology domain, it retains the residues that are phosphorylated by PDK1 (Thr256 and Ser422). PDK1 activates SGK in vitro by phosphorylating Thr256. Like Akt/PKB, SGK1 preferentially phosphorylates serine and threonine residues that lie in RXRXXS/T motifs. SGK1 can be activated by PI-3 Kinase, which makes it an important signaling molecule in growth factor and insulin dependent signaling pathways.

Applications
Suitable for use in Western Blot and Immunoprecipitation. Other applications have not been tested.
Recommended Dilution
Western Blot: 0.5-2ug/ml detects SGK1 in RIPA lysates from A431 and Rat 2 cells. Rat 2 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with S1010-83 (0.5ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunoprecipitation: 4ug Immunoprecipitates SGK1 from 500ug of Rat 2 lysate. Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recognizes human SGK1, Mr ~50kD. A protein, competed by the peptide, was also detected in Rat 2 lysates, Mr ~31kD.|Species Crossreactivity: rat.
