MaxLight™490 is a new Blue-Green photostable dye conjugate comparable to DyLight™488, Alexa Fluor™488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm); Emission (515nm); Extinction Coefficient 73,000.

SHP2 is a ubiquitously expressed tyrosine-specific protein phosphatase. It participates in signaling events downstream of receptors for growth factors, cytokines, hormones, antigens and extracellular matrices in the control of cell growth, differentiation, migration, and death. Activation of SHP 2 and its association with Gab1 is critical for sustained Erk activation downstream of several growth factor receptors and cytokines. In addition to its role in Gab1-mediated Erk activation, SHP 2 attenuates EGF-dependent PI3 kinase activation by dephosphorylating Gab1 p85 binding sites.
Applications
Suitable for use in Western Blot, Immunoprecipitation, Neutralization and Immunocytochemistry. Other applications have not been tested.
Recommended Dilution
Western Blot: detects SHP 2/SHPTP-2 in RIPA lysates from human A431 carcinoma cells and mouse 3T3/A31 fibroblasts. Strongly recognizes the SHP 2/SHPTP-2 fusion protein (10ng) used for immunoprecipitation; use 1ug/ml for this application. Immunoprecipitation: immunoprecipitates SHP 2/SHPTP-2 from RIPA lysates of human A431 carcinoma cells. An immunoprecipitation phosphatase assay using phenylphosphate as a substrate was performed using EGF stimulated A431 cells. Immunocytochemistry: detects SHP 2/SHPTP-2 in EGF stimulated A431 cells. Neutralization: Does not inhibit phosphotyrosine phosphatase activity. Optimal dilutions to be determined by the researcher.

A0001-50: Non stimulated A431 cell lysate.
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™490 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
