Mouse Anti-SOCS 1 (Suppressor of Cytokine Signalling 1, Janus kinase (JAK) binding protein, STAT-induced STAT inhibitor, SSI-1) (MaxLight 650)

MaxLight™ 650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000.

SOCS (Suppressor of Cytokines Signaling) proteins appear to form part of a classical negative feedback loop that regulates cytokine signal transduction. Transcription of each of the SOCS genes occurs rapidly in vitro and in vivo in response to cytokines, and once produced, the various members of the SOCS family appear to inhibit signaling in different ways. SOCS 1, also called Janus kinase (JAK) binding protein (JAB), or STAT-induced STAT inhibitor (SSI-1), is an SH2-domain-containing protein. It was shown to interact with all four Jak kinases (Jak1, Jak2, Jak3 and Tyk2) and is induced by and inhibits the signaling action of IL-3, IL-4, IL-6, growth hormone, LIF, prolactin, interferon gamma (IFNg), and erythropoietin (2). It plays an important role in the negative regulation of IFNg signaling and in the differentiation of T cells (3). SOCS 1 mRNA is highly expressed in the thymus, to a much lower level in the spleen, and not expressed in most other tissues (4). The SOCS 1 protein is primarily expressed in the thymocytes independent of the cytokine and T cell receptor (5). Biochemical characterization as well as gene disruption studies indicate that SOCS 1 is an important negative regulator of the JAK-STAT signal pathway.
Applications
Suitable for use in FLISA, Western Blot, Immunohistochemistry and Immunoprecipitation. Other applications not tested.
Recommended Dilutions
Immunohistochemistry (Paraffin): Requires HIER using sodium citrate buffer pH6.0 prior to staining. Optimal dilutions to be determined by the researcher.
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.

Specificity
Recognizes mouse SOCS 1, NT at ~30kD. Reactivity confirmed with mouse thymus, spleen, lung and liver homogenates as well as with mouse recombinant protein. Species Crossreactivity: human.
