Rabbit Anti-S-phase Kinase-associated Protein 2 (SKP2, Cyclin-A/CDK2-associated Protein p45, F-box Protein Skp2, F-box/LRR-repeat Protein 1, FBL1, FBXL1, FLB1, MGC1366, p45skp2)

The F box protein Skp2 (S-phase kinase-associated protein 2) is oncogenic, and its frequent amplification and overexpression correlate with the grade of malignancy of certain tumors. Skp2 controls p300-p53 signaling pathways in cancer cells, making it a potential molecular target for cancer therapy. This gene positively regulates the G(1)-S transition by controlling the stability of several G(1) regulators, such as the cell cycle inhibitor p27. This study provides evidence of a role for an F-box protein in oncogenesis and establishes SKP2 as a protooncogene causally involved in the pathogenesis of lymphomas.

Applications
Suitable for use in Western Blot, Immunohistochemistry and Immunocytochemistry. Other applications not tested.
Recommended Dilutions
Western Blot:1ug/ml Immunohistochemistry:1-2ug/ml detects SKP2 in formalin fixed and paraffin embedded tissues. Immunocytochemistry:1-2ug/ml detects SKP2 in acetone fixed cells. Antigen retrieval by pepsin and trypsin is required. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH20. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Synthetic peptide corresponding to a sequence from the N-terminal of human S-phase Kinase-associated Protein 2. Species Sequence Homology: rat and mouse (different by three amino acids)

Supplied as a lyophilized powder from PBS, 5% BSA, 0.05% thimerosal, 0.05% sodium azide. Reconstitute with 200ul sterile ddH2O.

Purified by immunoaffinity chromatography.
