Mouse Anti-Talin (KIAA1027, TLN, TLN1, ILWEQ)
Immunoblots the 225kD talin band on SDS gels of platelets and other cells. It may be used to stain adhesion plaques in fixed and permeabilised fibroblasts. T0750-20A recognizes an epitope within residues 139-433. This gene encodes a cytoskeletal protein that is concentrated in areas of cell-substratum and cell-cell contacts. The encoded protein plays a significant role in the assembly of actin filaments and in spreading and migration of various cell types, including fibroblasts and osteoclasts. It codistributes with integrins in the cell surface membrane in order to assist in the attachment of adherent cells to extracellular matrices and of lymphocytes to other cells. The N-terminus of this protein contains elements for localization to cell-extracellular matrix junctions. The C-terminus contains binding sites for proteins such as beta-1-integrin, actin, and vinculin.
Applications
Suitable for use in Western Blot, Flow Cytometry, Immunoprecipitation, Immunofluorescence and Immunohistochemistry. Other applications not tested.
Recommended Dilutions
Flow Cytometry: 1:25-1:100; 10ul labels 10e6 cells in 100ul. Membrane permeabilization is required. Immunohistochemistry: Frozen sections Optimal dilutions to be determined by the researcher.
Hybridoma
Spleen cells from immunized BALB/c mice were fused with cells of the SP2/0 mouse myeloma cell line.

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
