Rabbit Anti-TCF 1 (T-cell Factor 1)

LEF1/TCF are members of the high mobility group (HMG) DNA binding protein family of transcription factors which consists of four members: Lymphoid enhancer factor 1 (LEF1), T Cell Factor 1 (TCF1), TCF3 and TCF4. LEF1 and TCF1 were originally identified as important factors regulating early lymphoid development and are downstream factors in the Wnt signaling pathway. LEF1/TCF bind to Wnt response elements to provide a docking site for beta-catenin which translocates to the nucleus and promotes the transcription of target genes. LEF1/TCF proteins are dynamically expressed during development and aberrant activation of the Wnt signaling pathway is involved in many types of cancers such as colon cancers. TCF1, also known as Transcription Factor 7 (TCF7), has several isoforms due to alternative splicing and transcription from an alternative promoter. The isoforms generated by the alternative promoter do not contain the amino-terminal beta-catenin binding domain and therefore may function in a dominant negative manner. TCF1 displays dynamic expression both in the total amount and the type of isoforms expressed in T cells during development and differentiation.

Applications
Suitable for use in Flow Cytometry, Immunofluorescence, Immunocytochemistry, Immunohistochemistry, Immunoprecipitation and Western Blot. Other applications not tested.
Recommended Dilutions
Flow Cytometry: 1:100 Immunofluorescence (IC): 1:100 Immunohistochemistry (Paraffin): 1:100. Citrate unmasking Immunoprecipitation: 1:50 Western Blot: 1:1000. Incubate membrane with diluted antibody in 1X TBS, 5% BSA, 0.1% Tween-20 at 4°C with gentle shaking, overnight. Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Synthetic peptide corresponding to a region surrounding Pro95 of human TCF1 protein.

Supplied as a liquid in 10mM sodium HEPES, pH 7.5, 150mM sodium chloride, 0.1mg/ml BSA, <0.02% sodium azide, 50% glycerol.

Purified.
Specificity
Recognizes endogenous levels of total human TCF1 protein at 48 and 50kD. Does not recognize the dominant negative isoforms of TCF1 lacking the amino-terminal beta-catenin binding domain and does not crossreact with LEF1. Species Crossreactivity: mouse.
