Rat Anti-Uteroglobin Related Protein 1 (UGRP1, Secretoglobin Family 3A Member 2, Pneumo Secretory Protein 1, PnSP-1)

UGRP1 (also secretglobin gene family 3A2/SCGB3A2) is an 18 kD, secreted nonglycosylated polypeptide that is a member of the uteroglobulin family, UGRP subfamily. Mouse UGRP1(A) circulates as a 37 kD homodimer. Its mature form is 70 amino acids in length. There are two large alternate splice forms termed B and C. UGRP1B is a 113 aa secreted homodimer that shows a 22 aa insert between aa's 84 and 85 of the short form. UGRP1C theoretically shows a 55 aa substitution for the C-terminal seven aa of the short form. Its existence is unclear. Mature mouse UGRP1A is 86% and 81% aa identical to rat and human UGRP1, respectively.

Hybridoma
Produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a rat immunized with purified, E,coli-derived, recombinant mouse uteroglobin-related protein-1 (rmUGRP1; aa 22-139: Accession #Q920H1).
Applications
Western Blot: This antibody can be used at 1-2ug/ml with the appropriate secondary reagents to detect mouse UGRP1. Using a colorimetric detection system, the detection limit for rmUGRP1 is approximately 5 ng/lane under non-reducing and reducing conditions. Chemiluminescent detection will increase sensitivity by 5 to 50 fold. Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect mouse UGRP1. The detection limit for rmUGRP1 is approximately 25 ng/well. Optimal dilutions should be determined by each laboratory for each application.
Storage and Stability
Lyophilized powder may be stored at 4°C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Supplied as a lyophilized powder from a 0.2um filtered solution in PBS with 5% trehalose. Reconstitute with sterile PBS, 40-50% glycerol. If 0.2ml is used, the concentration will be 0.5mg/ml.

Specificity
Selected for its ability to detect rmUGRP1 in direct ELISAs and Western Blots. In these formats, this antibody shows no crossreactivity with rhUGRP1.
