Histone H3, dimethyl (Lys27) (APC)
Histone H3 is one of the 5 main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N terminal tail H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure. Acetylation of histone H3 occurs at several different lysine positions in the histone tail and is performed by a family of enzymes known as Histone Acetyl Transferases (HATs).
Applications
Suitable for use in Western Blot, Peptide Inhibition, Immunocytochemistry, Chromatin Immunoprecipitation, Beadlyte Histone-Peptide Specificity Assay and Dot Blot. Other applications not tested.
Recommended Dilution
Western Blot: 0.5ug/ml detects dimethylated histone H3 in acid extracted proteins from HeLa cells. No detection of 1ug unmethylated recombinant Histone H3 (H5110-12K) was observed. Peptide Inhibition: 1uM of the immunizing peptide to completely abolishes detection of histone H3 in Western Blot analysis of HeLa acid extracts. Slight signal reduction was observed with peptide containing dimethyl-lysine 9. Signal was not reduced by branched monomethyl H3(Lys27), linear monomethyl H3(Lys27), linear trimethylH3(Lys27), or linear dimethyl H3(Lys4) peptides. Beadlyte Histone-Peptide Specificity Assay: 12.5ng/ml or a range of dilutions detected peptides containing the dimethyl-lysine modification at residue 27. Peptides containing other modifications were not detected. Optimal dilutions to be determined by the researcher.
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
