MaxLight™ 650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000.

Tyrosinase is the principal enzyme required for the synthesis of melanin pigment by melanocytes in mammals. It is also a marker of melanocyte differentiation, and is commonly expressed by malignant melanoma. Tyrosinase immunoreactivity studies in formalin-fixed, paraffin-embedded tissues have demonstrated that the T311 antibody is a specific, sensitive marker for the detection of melanocytic lesions. Immunoreactivity has also been shown to correlate inversely with clinical stage of melanoma progression. Because tyrosinase is recognized by cytotoxic T cells from melanoma patients, it has been proposed as an immunotherapeutic target for melanoma vaccines. The molecular mass of tyrosinase is 60-75kD. However, the processed, glycosylated forms of the protein found in melanosomes have been reported at 70-75kD. The T311 antibody identifies a cluster of proteins at 70-80kD, consistent with previous reports.
Applications
Suitable for use in Immunohistochemistry and Western Blot. Other applications have not been tested.
Recommended Dilution
Immunohistochemistry: 1ug/ml Western Blot: 1-5ug/ml Note: Heat induced epitope retrieval (HIER) with EDTA, pH 8.0, prior to staining, is recommended for optimal immunohistochemical results with paraffin-embedded tissues. Optimal dilutions to be determined by the researcher.
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
