Specificity
Specific for the 4-5kD human b-Amyloid peptide derived by cleavage from the b-Amyloid precursor protein. Reactivity has been confirmed by Western Blot using a synthetic b-Amyloid peptide (1-40), by immunohistochemistry on Alzheimer’s diseased human brain sections and by immunoprecipitation of 35S-methionine labeled b-Amyloid peptide secreted by transfected culture cells.

MaxLight™ 650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000.

The progressive deposition of insoluble amyloid protein plaques in specific areas of the brain and its blood vessels is associated with a number of neuropathologies and is an early invariant feature of Alzheimer’s disease. The major component of these protein plaques is a peptide of 39-42 amino acids derived from a group of 695-770 amino acid precursor proteins collectively referred to as the b-amyloid precursor protein (Beta-APP). b-APP is highly conserved in vertebrates; however, its normal function has not been well defined. In each of the b-APPs, the b-Amyloid peptide (b-AP) is an internal peptide that begins about 99 residues from the carboxy-terminal end of the b-APP and extends from the extracellular domain into the middle of the hydrophobic, membrane spanning domain of the precursor protein. The precise mechanism whereby b-AP accumulates in brain remains poorly defined.
Applications
Suitable for use in FLISA, Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested.
Recommended Dilution
FLISA: 0.1-1ug/ml Western Blot: 1-3ug/ml Immunoprecipitation: 5-10ug; For immunoprecipitation of b-Amyloid peptide from cultured cells, it is necessary to mix the culture medium with an equal volume of 2X RIPA buffer. Immunohistochemistry: 5ug/ml Optimal dilutions to be determined by the researcher.
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
