This Phosphorylated Neurofilament H (pNF-H) BioAssay™ ELISA Kit is a sandwich ELISA for the in vitro quantitative determination of pNF-H concentrations in human serum, plasma and other biological fluids.
Detection Range
78.13-5000pg/ml
Precision
Intra-Assay %CV: <10% Inter-Assay %CV: <10%
Specificity
Recognizes human pNF-H. No significant cross-reactivity or interference between human pNF-H and analogues was observed.
Test Principle
The microtiter strips provided in this kit has been pre-coated with an antibody specific to pNF-H. Standards or samples are then added to the appropriate microtiter strip wells with a biotin-conjugated antibody specific to pNF-H. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Free components are washed away. The substrate solution is added, only those wells that contain pNF-H, biotin-conjugated antibody and enzyme-conjugated Avidin will apear blue in color. The enzyme-substrate reaction is terminated by the addition of sulfuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450nm ± 2nm. The OD value is proportional to the concentration of human pNF-H. The concentration of pNF-H in the sample is then determined by comparing the O.D. of the sample to the standard curve.
