Basal Medium Eagle (BME) developed by Harry Eagle, is one of the most widely used of all synthetic cell culture media. BME was developed a result of numerous studies carried out in the late 1950’s to determine the essential nutritional requirements and other factors critical to the growth of cells in culture. BME, when properly supplemented, has demonstrated wide applicability, for supporting monolayer growth of a wide variety of normal and transformed cell lines. BME is the predecessor of Eagle’s Minimum Essential Medium (MEM) and Dulbecco’s Modified Eagle’s Medium (DME). BME is suitable for use with diploid and primary mammalian cell cultures.
Appearance
White to off-white, homogenous, free flowing powder
Solubility (12.14g/L)
Clear, colorless, complete
Directions per Liter
Dissolve 12.14g in 800-900ml of ddH2O, stirring gently until completely solubilized. Add 2.2g sodium bicarbonate. Adjust pH of the medium to 0.1-0.3 pH unit below the desired level. Add additional water to bring the solution to 1L. Filter-sterilize using a 0.22 micron membrane filter. Aliquot into sterile containers. Do not autoclave. Contains heat-labile compounds that can be damaged with autoclaving.
Storage
Store powdered media at RT. Stable for 12 months after receipt. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time.
