Dulbecco’s Phosphate Buffered Saline (PBS) (10X) w/CaCl2, MgCl2 (Powder)

Dulbecco's Phosphate Buffered Saline (DPBS) is intended for use in the maintenance of mammalian cells where a chemically defined, balanced salt solution provides an environment that will maintain the structural and physiological integrity of cells in vitro. DPBS is commonly used in cell enumerations as a diluent, for rinsing cells and as a buffer in many chromotographic procedures. DPBS also is used to wash and resuspend cells during the dissociation process.

Solubility (1X): Colorless, clear, complete. Some precipitate may be observed as a 10X due to excess concentration of salts. Sterile filter after solubilization.

To make a 10X solution
Dissolve 97.8g in 800-900ml of ddH2O stirring gently until completely solubilized. Adjust pH of the medium to the desired level. Add additional water to bring the solution to1L Filter-sterilize using a 0.22 micron membrane filter. Aliquot into sterile containers..
Note: Some precipitate may be observed due to excess concentration of salts. Sterile filter after solubilization.
USB recommends the preparation of this product as a 1X solution.
To make a 1X solution
Dissolve 9.78g in 800-900ml of ddH2O stirring gently until completely solubilized. Adjust pH of the medium to the desired level. Add additional water to bring the solution to1L Filter-sterilize using a 0.22 micron membrane filter. Aliquot into sterile containers.
Storage and Stability
Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time.

To make a 10X solution
Dissolve 97.825 grams in ~950ml of ddH2O, stirring gently with heating until completely solubilized. Allow to cool and then adjust pH of the medium to the desired level. Add additional water to bring the solution to1L. Solution may be filter-sterilized or autoclaved for 15 minutes at 121ºC (15psi). Bottles of stored solution can be warmed in a hot water bath prior to use if crystallization is observed.
To make a 1X solution
Dissolve 9.782 grams per liter of distilled/deionized (DDI) water. Stir until completely dissolved. Adjust pH as necessary. May be filter-sterilized or autoclaved for 15 minutes at 121ºC (15psi).
USB recommends the preparation of this product as a 1X solution.
Storage and Stability
Store powdered media at RT. Stable for 12 months after receipt. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time.
