One of the most commonly used plant culture media, Murashige and Skoog Medium has proven effective in the culture of tissue derived from moncotyledons and dicotyledons. This medium, originally formulated to support tobacco callus, has been used to support callus initiation and growth, the growth of cells in suspension culture and the regeneration of shoots and plantlets from explants. Most recently, it has been used for micropropagation of ornamental, vegetable and fruit plants in research and commercial enterprises. Murashige and Skoog Salt Mixture contains all the basal salts as prescribed by Murashige and Skoog’s original formulation (1962). It is most commonly modified for use with specific plant culture applications.

Directions per Liter
Dissolve 4.328 grams in 800-900ml of ddH2O stirring gently until completely solubilized. Adjust pH of the medium to the desired level. Add additional water to bring the solution to1L Filter-sterilize using a 0.22 micron membrane filter and aliquot into sterile containers. In lieu of filter-sterilization, the solution can be autoclaved for 15 minutes at 121ºC (15psi).
Storage and Stability
Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time.
