RT/4°CNutrient Mixture Ham’s F-12 Kaighn’s Modification w/o Amino Acids (Powder)
Ham’s Nutrient Mixtures were originally developed to support clonal growth of several clones of Chinese hamster ovary (CHO) cells, as well as clones of HeLa and mouse L-cells. Both mixtures were formulated for use with or without serum supplementation, depending on the cell type being cultured. Ham’s F-10 has been shown to support the growth of human diploid cells, white blood cells for chromosomal analysis, primary explants of rat, rabbit and chicken tissues. Ham’s F-12 has been used for the growth of primary rat hepatocytes and rat prostate epithelial cells. May be used in combination with hormones and transferrin. For optimum buffering, use in a 5% CO2 atmosphere.

Directions per Liter
Dissolve 10.01g in 800-900ml of ddH2O stirring gently until completely solubilized. Add 1.18g/L sodium bicarbonate. Adjust pH of the medium to 0.1-0.3 pH unit below the desired level. Add additional water to bring the solution to1L. Filter-sterilize using a 0.22 micron membrane filter. Aliquot into sterile containers. Do not autoclave. Contains heat-labile compounds that can be damaged with autoclaving.
Storage and Stability
Store powdered media at RT. Stable for 12 months after receipt. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time.
