Sporulation Medium (Powder)

Standard medium used for inducing sporulation of yeast. Yeast cells will undergo several divisions on this medium. Sporulation occurs after 3-5 days of incubation. Nutrient suplements are required for sporulation of auxotrophic diploids on on Sporulation Medium. Supplement at the level of 25% of those used for most complete media should be added when componding the medium. Alternatively, supplements can be spread on teh surface of the Sporulation Medium plates. The liquid should be allowed to dry thoroughly onto the agar before inoculating with yeast strains.

Appearance
White to light tan, homogeneous, free flowing powder
Solubility
Colorless to light yellow, clear, complete with heat.
pH
As Reported
Directions per Liter
Dissolve 31.5 grams in 800-900ml of ddH2O stirring gently with heating until completely solubilized. Adjust pH of the medium to the desired level. Add additional water to bring the solution to1L. If required, add sodium bicarbonate. Dispense into appropriate containers, loosen caps and autoclave for 15 minutes at 121ºC (15psi).
Storage and Stability
Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept refrigerated and used within a short period of time.
