Plate Count Agar
A standard methods medium used in water, dairy products and food analysis.
Appearance: Light beige to tan, homogeneous, free-flowing powder
Solubility: Light beige to amber, clear to slightly hazy, complete after autoclaving
Final pH: 7.0 ± 0.2 at 25°C
Expected Cultural Response (18-24h at 35°C)
Lactobacillus acidophilus ATCC 11506: Good growth Staphylococcus aureus ATCC 25923: Good growth

Directions per Liter
Suspend 23.5g of the powder in 1L of distilled water. Mix until evenly dispersed. Heat with repeated stirring and boil for 1 minute to dissolve completely. Dispense into appropriate containers, loosen caps and autoclave for 15 minutes at 121ºC (15psi). Sterile solidified medium can only be remelted once; do not heat medium longer than 3 hours at 45-50ºC .
Storage and Stability
Store powdered media at RT. Stable for 12 months after receipt. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time.
