Terrific Broth, Modified for Genomics (Powder)

Terrific Broth is a highly enriched bacteriological medium specifically designed by Tartof and Hobbs to increase the yield of plasmid DNA derived from suspension cultures of E. coli due in part to the increased buffering capacity derived from using mono and dibasic phosphates. T2810-10 is useful for obtaining the highest yields of plasmid DNA from E.coli by the addition of a proprietary blend of vitamins, micronutrients and carbohydrates. Will produce up to 300% greater growth than traditional Terrific Broth!. Doubling time will actually decrease from 60 to 20 minutes, thus giving the same cell yield as Terrific Broth in 1/3 the time.

Directions per Liter
Dissolve 35 grams in 800-900ml of ddH2O stirring gently with heating until completely solubilized. Add additional water to bring the solution to 1L. Dispense into appropriate containers, loosen caps and autoclave for 15 minutes at 121ºC (15psi). If precipitate forms on autoclaving, medium may be filter-sterilized using a 0.2 micron filter.
Storage and Stability
Store powdered media at RT. Stable for 12 months after receipt. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept refrigerated and used within a short period of time.
