The chitinolytic enzymes from T. viride are a mixture of extracellular chitinolytic enzymes, which exhibit exo and endochitinase activities including N-acetyl-β-glucosaminidase and chitobiosidase.

Chitinases are digestive enzymes that break down glycosidic bonds in chitin. Because chitin composes the cell walls of fungi and exoskeletal elements of some animals (including worms and arthropods), chitinases are generally found in organisms that either need to reshape their own chitin or to dissolve and digest the chitin of fungi or animals.

Chitinase Unit Definition
One unit will liberate 1mg of N-acetyl-D-glucosamine from chitin per hour at pH 6.0 at 25°C in a 2 hour assay.
Chitobiosidase Unit Definition
N-Acetyl-b-glucosaminidase [Chitobiosidase]: One unit will release 1.0mg of 4-nitrophenol from 4-nitrophenyl N-Acetyl-b-Dglucosaminide [4-nitrophenyl N,N'-diacetyl -b--Dchitobioside] per minute at pH 6.1 at 40°C.
Assay
Reaction mixture is composed of 30mg of N-acetyl chitosan, 2ml of 0.1M Mcllvaine buffer (pH 6.8) and 1ml of enzyme solution. After incubation at 37°C for 60 minutes, the reaction of the enzyme is terminated by heating at 100°C for 5 minutes and the amount of reducing sugar is determined by the method of Schales and Schales, using N-acetyl glucosamine as a standard.
Storage and Stability
Lyophilized powder may be stored at 4°C for short-term only. Stable at -20ºC for 6 months after receipt. Reconstitute with sterile PBS. Store at -20°C. Reconstituted product is stable for 5 weeks at 4°C. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
