Orexin Prepro, Control Peptide (Hypocretin Prepro)

Control Peptide for O7502 (antiserum) and O7502-05 (affinity purified)

Synthetic peptide, 13aa, murine Orexin A Pre-pro immunogenic peptide shows 82% (14/17 aa) and 70% (12/17 aa) identity with rat and human prepro-orexins, respectively. The sequence is not found in Orexin A or Orexin B. Therefore, antibodies will not detect mature Orexin A or Orexin B. Use the appropriate control immunogenic peptides to confirm specificity of antibodies.
Hypothalamic neuropeptides associated with feeding behavior, binding and activating two closely related Orexin receptors—G-protein coupled receptors (GPCRs) OX1R and OX2R. Recognizes the full-length (1-33 aa) Orexin-A. The epitope is not found in Orexin B. Since Orexin A and Orexin B are derived from an unprocessed prepro-orexin (130 aa), anti-Orexin A antibodies may also react with pre-pro orexins.
Applications
Suitable for use in ELISA and Antibody Blocking. Not suitable for use in Western Blot due to low molecular weight. Other applications not tested.
Recommended Dilution
ELISA: 50-100ng control peptide/well. Antibody Blocking: 5-10ug per 1ug O7502-05 (affinity purified antibody) or per 1ul O7502 (antiserum). Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile buffer or ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
