Brain Heart Infusion Broth without Dextrose is a modification of the media described by Rosenow and Hayden in which infusion from calf brains has replaced the brain tissue and disodium phosphate has replaced the calcium carbonate buffer. The addition of 0.1% agar can be used to lower oxygen tension to enhance the growth of anaerobic organisms. Brain Heart Infusion Broth without Dextrose is a basal medium used with added carbohydrates for fermentation studies.
Dissolve 35g in 800-900ml of ddH2O stirring gently with heating until completely solubilized. Adjust pH of the medium to the desired level. Add additional water to bring the solution to1L. Dispense into appropriate containers, loosen caps and autoclave for 15 minutes at 121ºC (15psi).
Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept refrigerated and used within a short period of time.
