Dextrose agar is used for the culture of microorganisms with or without added blood. Supplementation with 5% blood provides additional growth factors for fastidious microorganisms. When pH is lowered to 3.5 medium can be used to obtain total counts of yeasts in food.
Suspend 43g of the powder in 1L of distilled water. Mix thoroughly. Heat with frequent agitation and boil for 1 minute to completely dissolve the powder. Dispense into appropriate containers, loosen caps and autoclave for 15 minutes at 121ºC (15psi).
Optional: To prepare blood agar, aseptically add 5% sterile defibrinated blood to the medium at 45-50°C. Mix well and dispense as desired.
Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept refrigerated and used within a short period of time.
