DNase Test Agar with Toluidine Blue is a complete medium since toluidine blue is incorporated in the formulation. A positive reaction is indicated by a rose-pink zone surrounding the colony in a dark blue background. In general, Gram positive microorganisms are inhibited. This medium is particularly useful for determining DNase activity of the Enterobacteriaceae.
Dissolve 42grams per liter of distilled/ deionized (DDI) water, heating with stirring until completely solubilized.
Dispense into appropriate containers, loosen caps and autoclave for 15 minutes at 121ºC (15psi)
Store powdered media at 4°C Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should stored at 4°C and used within a short period of time.
